Principle / Procedure
The isolation of genomic DNA (gDNA) from the majority of organs is a challenging task. Organs are composed of different tissues with various types of cells and matrices. Large molecules such as collagens and proteoglycans can impede lysis and even impair downstream enzymatic reactions (e.g., PCR). The new NucleoSpin® 96 DNA RapidLyse kit uses a unique lysis chemistry to release gDNA from organs and cells with the highest efficiency.

Highest DNA Yields
Different mammalian organs were lysed and DNA was extracted. Same amounts of individual eluates were subjected to gel electrophoresis. Highest yields of DNA could be extracted with NucleoSpin® 96 DNA RapidLyse. Q represents a competitor kit, MN represents NucleoSpin® 96 DNA RapidLyse.
Best qPCR performance
DNA was isolated with NucleoSpin® 96 DNA RapidLyse (orange) or with a kit from Competitor Q (blue). A 136 bp fragment of the 18S rRNA gene was amplified by qPCR. Lowest Ct-values were detected when DNA was isolated with NucleoSpin® 96 DNA RapidLyse, indicating highest gDNA yields.
