Product Class: Restriction Endonuclease

MmeI
neb31 cloned at NEB recombinant time saver 2site dil_B 37 65 Heat CpG

SAM now included in enzyme formulation – no longer supplied as a separate vial.

We are excited to announce that all reaction buffers are now BSA-free. NEB began switching our BSA-containing reaction buffers in April 2021 to buffers containing Recombinant Albumin (rAlbumin) for restriction enzymes and some DNA modifying enzymes. Find more details at www.neb.com/BSA-free.

NEB restriction endonuclease that recognizes the sequence TCCRACNNNNNNNNNNNNNNNNNN_NN^

Product Introduction

  • Time-Saver™ qualified for digestion in 5-15 minutes
  • 100% activity in rCutSmart Buffer (over 210 enzymes are available in the same buffer) allowing for easier double digests
  • Type IIS restriction enzymes recognize asymmetric DNA sequences and cleave outside of their recognition sequence
  • Requires two or more sites for cleavage. Please review the enzyme list and recommendations in the following table as well as the background information article.
  • Restriction Enzyme Cut Site: TCCRAC(20/18)

FAQs & Troubleshooting

FAQs

  1. Is there any variability in the distance of cutting from the MmeI cut site which is indicated as 20/18?
  2. I am observing partial digestion when using MmeI. What could be the reason?
  3. Is MmeI sensitive to CpG methylation?
  4. Why can a cleaved MmeI site religate but not be re-cut?
  5. Is MmeI cutting exact or is the spacing variable?
  6. Do I have to set-up digests with Time-Saver™ qualified enzymes for 5-15 minutes? Can I digest longer?
  7. I tested your restriction enzyme on the substrate DNA recommended by NEB, and it appears to be active, however it does not digest my DNA. What could be the reason?
  8. Will incubation overnight improve my digest?
  9.  Does MmeI require SAM for activity?
  10. Can you tell me more about the switch from BSA to Recombinant Albumin (rAlbumin) in NEBuffers?
