This peptide is used to block Bcl-xL (54H6) Rabbit mAb #2764 and Bcl-xL Antibody #2762 reactivity.
The quality of the peptide was evaluated by reversed-phase HPLC and by mass spectrometry. The peptide blocks Bcl-xL (54H6) Rabbit mAb #2764 and Bcl-xL Antibody #2762 signal in both immunohistochemistry and western blotting.
Western Blot analysis of extracts from Ramos and Raw cells using Bcl-xL (54H6) Rabbit mAb #2764 (left) or the same antibody preincubated with Bcl-xL Blocking Peptide (right).
Western Blot analysis of extracts from Ramos and Raw cells using Bcl-xL Antibody #2762 (left) or the same antibody preincubated with Bcl-xL Blocking Peptide (right).
Immunohistochemical analysis of paraffin-embedded human lung carcinoma using Bcl-xL Antibody #2762 in the presence of control peptide (left) or Bcl-xL Blocking Peptide (right).
Directions For Use
Use as a blocking reagent to evaluate the specificity of antibody reactivity in western immunoblotting and immunohistochemistry protocols. For Immunohistochemistry, add twice the volume of peptide as volume of antibody used in 100 µl total volume. Incubate for a minimum of 30 minutes prior to adding the entire volume to the slide. For western blotting, add an equal volume of peptide as volume of antibody used in 10 ml total volume, and incubate at room temperature for 30 minutes before allowing to react with the blot. Recommended antibody dilutions can be found on the Bcl-xL Antibody #2762 and Bcl-xL (54H6) Rabbit mAb #2764 data sheets.
Bcl-xL prevents apoptosis through two different mechanisms: heterodimerization with an apoptotic protein inhibits its apoptotic effect (1,2) and formation of mitochondrial outer membrane pores help maintain a normal membrane state under stressful conditions (3). Bcl-xL is phosphorylated by JNK following treatment with microtubule-damaging agents such as paclitaxel, vinblastine and nocodazole (4,5).
