The expansion of flow cytometry applications from cellular analysis to molecular and genomic analysis, and the increase in availability of monoclonal antibodies and fluorochromes have magnified the demand for flow cytometers with expanded fluorescence detection channels. ACEA NovoCyte 3000VYB (405 nm, 488nm, 561nm) and NovoCyte 3000RYB (488nm, 561nm, 640nm) flow cytometers configured with 561nm laser are developed to meet the needs of multicolor analysis, broadening application capabilities in both basic research and clinical studies.
The advantages of using 561nm laser configured ACEA NovoCyte 3000VYB and NovoCyte 3000RYB flow cytometers include: enhancing fluorescent signals of PE-conjugated antibodies with 561nm excitation and increasing sensitivity on detecting dim signals; eliminating or minimizing FITC to PE spillovers by independent excitation with 488nm and 561nm lasers; favoring the detection of multiple fluorescent proteins such as GFP, DsRed and mCherry; fulfilling multicolor analysis of variety of applications.
Optical configurations of NovoCyte 3000 VYB, NovoCyte 3000 RYB and NovoCyte 3000 are listed in this application note. All the data in this application note by 561nm laser excitation were obtained from NovoCyte 3000 VYB or NovoCyte 3000 RYB flow cytometers, and data by 488nm laser excitation were from NovoCyte 3000 flow cytometer.
